Procedure for embryonic cell culture. (A) Perform this procedure under aseptic conditions. (B) Removing solution with yellow tip-capped aspiration needle (such as step 4). (C) Adding solution to 96-well plate after splitting embryos to each well (step 8). (D) Transferring embryo from 12-well plate to P10 dish using sterilized forceps (step 10). (E and F) Steps 11 to 13, where embryo is picked out aseptically, are performed under stereomicroscope using forceps. (G) The embryo picked out and transferred is dispersed by repetitively passing through 22 G needle (step 14). (H) After plating dispersed cells, cells should be kept at 27.5°C (step 15).
